Target Information
ICAD (Inhibitor of Caspase-3-Activated DNase) controls nuclear apoptosis by inhibiting CAD.
ICAD has two isoforms:
- ICAD-L/DFF45 (45 kDa) – functional isoform
- ICAD-S/DFF35 (35 kDa) – non-functional isoform
ICAD is cleaved by caspase-3 at residues 117 and 224, releasing CAD and initiating apoptotic DNA fragmentation.
In non-apoptotic cells, CAD is complexed with ICAD. ICAD also acts as a CAD-specific folding chaperone.
Recent studies suggest ICAD may be a potential target for restoring normal apoptotic signaling in colon and brain cancer cells.
Two alternatively spliced transcript variants encoding distinct isoforms have been identified for the ICAD gene.
