Target Information
The Lipin family of nuclear proteins includes Lipin-1, Lipin-2, and Lipin-3, all containing a nuclear localization signal, a conserved N-terminal (NLIP) domain, and a C-terminal (CLIP) domain.
LPIN1 (Lipin-1) is essential for normal adipose tissue development and metabolism. It activates PGC-1A target pathways, including fatty acid oxidation and mitochondrial oxidative phosphorylation, by inducing expression of the nuclear receptor PPARA.
LPIN1 also suppresses lipogenesis and circulating lipid levels. Overexpression promotes fat accumulation and insulin sensitivity, while deficiency can lead to lipodystrophy, fatty liver, hypertriglyceridemia, and insulin resistance.
