Product Specific Information
The HI30 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed on all hematopoietic cells except circulating erythrocytes and platelets. It plays a crucial role in lymphocyte activation through its tyrosine phosphatase activity.
- Applications Reported: Flow cytometric analysis
- Applications Tested: Pre-diluted and tested by flow cytometry of normal human peripheral blood cells
- Recommended Usage: 5 µL (0.25 µg) per test (100 µL final volume; 10⁵–10⁸ cells/test)
Fluorochrome Information:
Brilliant Violet™ 421 (BV421) emits at 423 nm and is suitable for cytometers with a violet (405 nm) laser. Ensure instrument compatibility prior to use.
Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies (e.g., Brilliant Violet™, Brilliant UV™, Super Bright™), use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Target Information
CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells (~10% of lymphocyte surface proteins). It is absent in non-hematopoietic cells and tissues. CD45 is essential for T- and B-cell receptor signaling, cytokine receptor regulation, and plays roles in apoptosis and integrin-mediated signal transduction. Multiple isoforms exist due to alternative splicing, differing in extracellular domains.
