Target Information
TNK2 (ACK), also known as activated Cdc42-associated kinase, is a non-receptor tyrosine kinase that functions downstream of Cdc42. It is implicated in tumorigenesis and metastasis, particularly in prostate cancer. TNK2 mediates CDC42-dependent cell migration via phosphorylation of BCAR1 and may play roles in synaptic function and brain development.
Key functions include:
- Activation of AKT1 via phosphorylation on Tyr-176
- Phosphorylation of AR on Tyr-267 and Tyr-363, enhancing recruitment to androgen-responsive enhancers (AREs)
- Phosphorylation of WWOX on Tyr-287 and MCF2 to enhance GEF activity
- Regulation of AXL receptor levels and modulation of tumor growth through interaction with tumor suppressors and pro-survival factors
