Product Specific Information

The eBio1A6 monoclonal antibody reacts with human thymic stromal-derived lymphopoietin receptor (TSLPR), a ~50 kDa protein similar to the common gamma-chain.
TSLPR complexes with IL-7R alpha (CD127) to form a high-affinity receptor binding TSLP. It is expressed by monocytes and CD11c+ dendritic cells. TSLP binding induces expression of Th2 cell-attracting chemokines CCL17 and CCL22, and activates dendritic cells to increase secretion of IL-4, IL-5, and IL-13.

In mice, TSLPR deficiency alone does not affect lymphocyte numbers, but double deficiency with the common gamma-chain reduces lymphocyte counts.
The eBio1A6 antibody cross-blocks binding of another anti-human TSLPR antibody, 1D8.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on TSLPR-transfected cells at 5 µL (0.5 µg)/test.
A test is defined as the amount needed to stain a sample in 100 µL final volume, for 10⁵–10⁸ cells/test.

Fluorophore Information:
Super Bright 436 is excited at 405 nm and emits at 436 nm.
Recommended filter: 450/50 bandpass or equivalent.

When using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.

Target Information

Thymic stromal lymphopoietin (TSLP) is a key cytokine driving dendritic cell maturation and activation. It supports regulatory T cell selection, CD4+ T cell homeostasis, and allergic response induction.
TSLP also promotes B cell progenitor growth and differentiation to IgM+ stage.
Its receptor (TSLP-R) binds TSLP with low affinity, but co-expression with IL-7Rα forms a high-affinity functional complex requiring STAT5 for signal transduction, inhibited by SOCS-1.
