Target Information
Crystallins are separated into two classes: taxon-specific (enzyme) and ubiquitous. The ubiquitous class constitutes the major proteins of the vertebrate eye lens and maintains lens transparency and refractive index. Since lens fiber cells lose nuclei during development, crystallins are synthesized and retained throughout life, making them extremely stable proteins.
Mammalian lens crystallins are divided into alpha, beta, and gamma families; beta and gamma crystallins form a superfamily. Gamma-crystallins are highly symmetrical, monomeric proteins lacking connecting peptides and terminal extensions. They are differentially regulated after early development. Four gamma-crystallin genes (gamma-A through gamma-D) and three pseudogenes (gamma-E, gamma-F, gamma-G) are organized in a genomic cluster. Aging or mutations in these genes are implicated in cataract formation.
