Product Specific Information

The eBio1D4B monoclonal antibody reacts with mouse CD107a (LAMP-1), a type I lysosomal membrane protein highly glycosylated and expressed in late endosomes-lysosomes. It is transiently expressed on the surface of degranulating cytolytic T cells and implicated in cancer metastasis and lysosomal storage disorders.

Applications Reported: Suitable for intracellular staining followed by flow cytometric analysis.
Applications Tested: Verified using mouse thioglycolate-elicited peritoneal exudate cells with the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00). Recommended usage ≤1.0 µg/test (100 µL final volume). Optimal titration required.

Fluorochrome Details:
Brilliant Violet™ 711 (BV711) emits at 713 nm and is intended for use on cytometers equipped with a violet (405 nm) laser. Ensure instrument compatibility.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to minimize non-specific interactions.

Handling Notes:

  • Light sensitive: protect from light.
  • Fixation: Compatible with IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C.
  • Not compatible with methanol-based fixation.

Target Information

LAMP1 (CD107a) is a major lysosomal membrane component alongside LAMP-2. It is a heavily glycosylated type I integral membrane protein with 18 N-linked glycosylation sites and a short cytosolic tail. LAMP1 is essential for lysosome-phagosome fusion and lysosome biogenesis.

Upon cell activation, LAMP1 translocates to the plasma membrane via a tyrosine-based motif (YXXI). Mutations affecting this motif disrupt lysosomal localization. Surface LAMP1 promotes PBMC adhesion to vascular endothelium, suggesting involvement in inflammation. Elevated LAMP1 levels are observed near senile plaques in Alzheimer’s disease and serve as a marker for mast cell activation.
