Target Information

Tyrosine hydroxylase (TH) is an enzyme involved in the synthesis of catecholamine neurotransmitters dopamine, epinephrine, and norepinephrine. Catecholamine synthesis is regulated by the interaction of TH with the cofactor tetrahydrobiopterin (BH4), which binds to the TH catalytic domain and activates enzymatic function.
In humans, four TH mRNA splice variants (hTH1–hTH4) have been identified, differing in their N-terminal regulatory domains but sharing identical catalytic domains.
TH catalyzes the conversion of L-tyrosine to L-dopa and plays a critical role in adrenergic neuron physiology. Dysregulation of TH is associated with several neuropathogenic diseases, including Parkinson’s disease, schizophrenia, Segawa syndrome, dystonia, and cardiovascular disorders.
