Target Information
Cytokine receptor signaling is negatively regulated by a family of Src homology 2 (SH2) domain-containing adaptor molecules termed SOCS (Suppressor of Cytokine Signaling). There are eight known members: SOCS-1 to SOCS-7 and CIS. These proteins share a structure consisting of a variable N-terminal region, a central SH2 domain, and a C-terminal SOCS box motif. SOCS proteins function as part of a negative feedback loop that regulates cytokine signal transduction via STAT-induced transcriptional mechanisms. Transcription of SOCS genes is rapidly induced by cytokines, and each member modulates signaling in distinct ways.
