SMAD 단백질은 Drosophila의 mothers against decapentaplegic (Mad) 및 C. elegans의 Sma 유전자 산물과 유사한 단백질 계열에 속합니다.
이 단백질은 신호전달 및 전사 조절에 관여하며, bone morphogenetic proteins (BMPs) 신호를 매개합니다.
BMP 리간드 자극 시, SMAD1은 BMP 수용체 키나아제에 의해 인산화되어 활성화되고, SMAD4와 복합체를 형성하여 전사 조절 기능을 수행합니다.
