The Non-Human Primate MCP-1 (CCL2) Simplex ProcartaPlex Kit measures MCP-1 (CCL2) protein and is designed to be combinable with other Simplex kits, allowing users to create custom multiplex panels utilizing Luminex xMAP technology for protein detection and quantitation. When combining multiple Simplex kits (not using a pre-configured Multiplex Panel), only one buffer kit (sold separately) is needed for each assay plate regardless of plex size.

About ProcartaPlex Assays for the Luminex Platform

ProcartaPlex immunoassays are based on the principles of a sandwich ELISA, using two highly specific antibodies that bind to different epitopes of a single protein. This enables simultaneous quantitation of multiple protein targets using a Luminex instrument. The assay requires as little as 25 μL of plasma or serum, or 50 μL of cell culture supernatant, and provides results within four hours.

ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for simultaneous detection and quantitation of up to 80 protein targets in a single small-volume sample from plasma, serum, cell culture supernatants, and other biological fluids.

The Luminex beads in the ProcartaPlex assay are internally dyed with precise proportions of red and infrared fluorophores to create spectrally unique signatures identifiable by Luminex xMAP detection systems (e.g., Luminex 200, FLEXMAP 3D, MAGPIX). Similar to a sandwich ELISA, the assay uses matched antibody pairs to identify target proteins. Each spectrally unique bead is labeled with antibodies specific for a single protein, and bound proteins are detected with biotinylated antibodies and streptavidin–R-phycoerythrin (RPE). This bead-based design allows analysis of multiple targets in a single well.

Unlike traditional ELISA, the capture antibody in a ProcartaPlex assay is conjugated to a bead rather than adsorbed to a microplate well, enabling reagents to remain free-floating in solution. The Luminex 200 instrument uses two lasers: one to distinguish bead spectral signatures and another to quantify RPE fluorescence, which correlates with protein concentration. ProcartaPlex assays allow profiling of more targets using less sample and time compared to conventional ELISA.
