Target Information
Nerve growth factor (NGF) influences the survival and differentiation of various neuronal and nonneuronal cells. Its biological activity results from binding to specific cell surface receptors, present as high and low affinity sites. Only the high affinity form mediates signal transduction. Cross-linking studies with ¹²⁵I-NGF show that the high affinity receptor of rat PC12 cells forms a 158 kDa complex with NGF, while the low affinity receptor forms a 100 kDa complex. Human and rat NGF receptor genes exhibit over 90% homology. NGF stimulates growth and differentiation of sympathetic and sensory neurons and is involved in their regulation. Mutations in the NGF gene are linked to hereditary sensory and autonomic neuropathy type 5 (HSAN5), and dysregulation of NGF expression is associated with allergic rhinitis.
