Target Information
The proteasome is a multicatalytic proteinase complex with a highly ordered ring-shaped 20S core structure, composed of four rings of 28 non-identical subunits (two alpha and two beta rings). Proteasomes are abundant in eukaryotic cells and cleave peptides in an ATP/ubiquitin-dependent, non-lysosomal pathway.
The immunoproteasome plays an essential role in processing class I MHC peptides. The PSMB8 gene encodes a member of the proteasome B-type family (T1B family), a 20S core beta subunit located in the MHC class II region. Expression is induced by gamma interferon, replacing the catalytic subunit 3 (proteasome beta 5 subunit) in the immunoproteasome. Proteolytic processing is required to generate a mature subunit. Two alternative transcripts encoding isoforms have been identified, both yielding the same mature subunit.
