Target Information
FAS (Fas, CD95, APO-1) is a 46 kDa transmembrane glycoprotein functioning as a cell death receptor in the TNFR superfamily. A soluble form (~26 kDa) also exists. Upon stimulation, FAS aggregates its intracellular FAS-associated death domains (FADD), forming the death-inducing signaling complex (DISC) that activates caspases. These caspases trigger a downstream cascade leading to apoptosis. FAS can activate NF-kappaB, MAPK3/ERK1, and MAPK8/JNK, participating in proliferative signaling in fibroblasts and T cells. Multiple alternatively spliced transcript variants encode distinct isoforms, with soluble forms potentially regulating apoptosis negatively. The Fas/Fas ligand system is implicated in diseases such as AIDS, hepatitis, and cancer, and plays a key role in antiviral immune responses.
