Target Information
FAS (Fas, CD95, APO-1)는 TNFR(superfamily)에 속하는 46 kDa 막단백질로, 세포 사멸 수용체로 작용합니다. FAS는 약 26 kDa의 가용성 형태로도 존재하며, FAS 자극은 FADD(FAS-associated death domain)와 DISC(death-inducing signaling complex) 형성을 유도하여 caspase 활성을 유도합니다. Caspase의 자동분해 과정은 세포 사멸(apoptosis)을 유도하는 하위 신호 전달을 촉발합니다. 또한 FAS는 NF-κB, MAPK3/ERK1, MAPK8/JNK를 활성화하며, 정상 섬유아세포 및 T세포의 증식 신호 전달에도 관여합니다. 최소 8개의 대체 스플라이싱 전사체가 보고되었으며, 막구역이 결여된 isoform은 완전 길이 isoform의 세포 사멸을 음성 조절할 수 있습니다. Fas/Fas ligand 시스템은 AIDS, 간염, 암 등 다양한 인체 질환과 관련되어 있으며, FAS ligand 경로를 통한 세포 사멸 유도는 항바이러스 면역 반응에 중요한 역할을 합니다.
