Target Information

PROZ (Protein Z) is a vitamin K-dependent plasma glycoprotein, approximately 62 kDa and 396 amino acids in length. It contains a Gla-rich region, two EGF-like domains, and a trypsin-like domain but lacks the catalytic serine residue typical of active serine proteases.
Protein Z participates in the coagulation cascade and is structurally related to factors VII, IX, X, and protein C. It binds to phospholipid surfaces via vitamin K-dependent carboxyglutamate residues. Its primary role is to facilitate the degradation of factor Xa by protein Z-related protease inhibitor (ZPI), enhancing the reaction rate up to 1000-fold. Deficiency in PROZ may be associated with either thrombotic or bleeding tendencies, though its physiological role is inhibitory.
