Product Specific Information

Description:
The HI100 monoclonal antibody reacts with human CD45RA, a 220 kDa molecule expressed by subpopulations of CD4+ and CD8+ peripheral T lymphocytes, as well as B cells. CD45RA+ T cell populations are mainly naïve/virgin, allowing the use of HI100 mAb as a phenotypic marker to discriminate T cell subsets.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use is 20 µL (0.008 µg) per test. A test is defined as the amount of antibody required to stain a cell sample in a final volume of 100 µL. Cell number can range from 10⁵ to 10⁸ cells/test.

Target Information

CD45RA is an isoform of the CD45 complex, expressed in specific lymphoid cell subtypes. CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase found on all nucleated hematopoietic cells, making up about 10% of lymphocyte surface proteins. It plays a key role in lymphocyte development, antigen signaling, and regulation of Src-family kinases.
CD45 isoforms differ due to alternative splicing, affecting extracellular domains while sharing identical transmembrane and cytoplasmic regions. Expression varies by cell type and physiological state. CD45 also contributes to integrin-mediated signaling and apoptosis regulation.
CD45 is expressed on all hematopoietic lineage cells except mature red cells and is absent in non-hematopoietic tissues. Dysregulation of CD45 is associated with severe combined immunodeficiency and Hepatitis C.
