Target Information
Estrogen Receptors (ER) belong to the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor acts as a ligand-activated transcription factor; upon binding to estrogen, it undergoes a conformational change that enables dimerization and binding to estrogen response elements (ERE) in DNA. This interaction allows ER to regulate gene transcription through coactivator or corepressor recruitment.
Two distinct forms exist—ER alpha and ER beta—encoded by ESR1 and ESR2 genes, respectively. Alternative RNA splicing generates at least four ER alpha isoforms (66 kDa, 53 kDa, 47 kDa, and 35 kDa). Estrogen receptors are expressed in various tissues, playing critical roles in sexual development, reproduction, bone maintenance, and are implicated in diseases such as breast and endometrial cancers, and osteoporosis.
