이 항체는 아세톤 고정된 동결절편 또는 세포 도말에 사용 가능하며, 파라핀 절편에는 적합하지 않습니다. FACS 분석에서 1차 항체로 사용되며, BMS121FI는 직접 염색에, BMS121BT는 biotin-(strept)avidin 시스템을 이용한 2단계 염색에 사용됩니다. ELISA에도 적합하나 기능적 분석에는 테스트되지 않았습니다.
Leukocyte-Endothelial Cell Adhesion Molecule-1은 selectin 계열의 접착 분자로, E-selectin, P-selectin, L-selectin과 함께 백혈구와 내피세포의 초기 상호작용을 매개합니다. L-selectin은 염증 부위로의 백혈구 이동을 유도하며, 음전하를 띤 리간드(시알산, 황산, 만노스, 푸코스 등)와 결합합니다. 또한 L-selectin은 활성화된 내피세포의 E-selectin과도 상호작용할 수 있습니다.
L-selectin은 대부분의 백혈구에서 발현되며, 림프구의 림프절 고내피 venule 결합과 호중구의 염증 부위 침투에 필수적입니다. 호중구 활성화 시 L-selectin은 막 근처에서 단백질 분해에 의해 절단되어 shedding되며, 림프구와 단핵구에서도 느리게 shedding이 일어납니다. Shed된 sL-selectin은 기능적으로 활성 상태를 유지하며, 높은 농도에서는 백혈구의 내피 부착을 억제할 수 있습니다. 혈청 내 sL-selectin의 주요 공급원은 조직 내 백혈구입니다.
Target Information
Human CD62L은 74–95 kDa의 당단백질로, selectin 계열 접착 수용체에 속합니다. 구조적으로 C-type lectin 결합 도메인, EGF-like 도메인, 두 개의 SCR 반복 서열, spacer 영역, 막관통 영역, 짧은 세포질 영역으로 구성됩니다. CD62L은 대부분의 백혈구(기억 T세포 일부 제외), 단핵구, 다형핵세포에서 지속적으로 발현됩니다.
