Target Information
Calpain-6, also known as calpamodulin or calpain-X, is an intracellular, calcium-dependent cysteine protease. Unlike the ubiquitous calpain-1 and calpain-2, calpain-6 shows tissue-specific expression, primarily in the placenta and embryonic tissues.
The calpain family consists of a small common subunit (calpain-4) and a large variable subunit. However, it is unclear whether calpain-6 associates with a small subunit. The large subunit includes domains I–IV, but calpain-6 lacks the EF-hand calcium-binding motif in domain IV.
Additionally, the canonical active site residues (Cys, His, Asn) are modified (Lys, His, Asn in human; Lys, Tyr, Asn in mouse), suggesting calpain-6 is not proteolytically active.
