Product Specific Information
The eBio4KB128 monoclonal antibody recognizes human CD22 (Siglec-2), a member of the siglec subgroup of the Ig superfamily.
CD22 is a type I transmembrane glycoprotein composed of two polypeptide chains (130 and 140 kDa) produced by alternative splicing.
It is highly expressed on mature B cells and B cell lymphomas.
The extracellular portion contains seven Ig-like domains capable of binding sialic acid moieties on epithelial, endothelial, B, and T cells.
The intracellular domain includes six tyrosine residues within ITIM and activation-like motifs, phosphorylated upon B-cell receptor engagement, enabling regulation of B-cell receptor signaling.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.06 µg) per test (100 µL final volume).
Cell number range: 10⁵–10⁸ cells/test.
Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm).
Recommended bandpass filter: 710/50; alternative: 695/40. Ensure instrument compatibility.
Fixation:
Samples may be stored in IC Fixation Buffer (Product # 00-822-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information
CD22 (BL-CAM) is a type I integral membrane glycoprotein (130–140 kDa) expressed in cytoplasm and membrane of B-lymphocytes.
It appears early in B-cell differentiation, similar to CD19 antigen.
Membrane expression is limited to mature B cells (CD22+) and absent in plasma cells (CD22−), making it valuable for phenotyping mature leukemias.
Strongly expressed in hairy cell leukemia, diffuse large B-cell lymphoma, and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.
CD22 preferentially binds α2,6-linked sialic acid.
Ligand-induced tyrosine phosphorylation participates in B-cell receptor signaling regulation, acting both as positive and inhibitory receptor through recruitment of phosphatases.
