Target Information
CD20 is a non-glycosylated surface phosphoprotein (33–37 kDa) expressed on mature and malignant B cells, a subset of T lymphocytes, and follicular dendritic cells. It regulates transmembrane calcium conductance, cell cycle progression, and B-cell proliferation.
CD20 associates with lipid rafts, with its intensity depending on extracellular triggers such as conformational changes or BCR aggregation. Upon receptor ligation, CD20 and BCR colocalize and then separate before endocytosis, with CD20 remaining on the cell surface.
CD20 is a key therapeutic target for antibody-mediated B-cell depletion in malignancies, as it remains membrane-bound after monoclonal antibody treatment.
Diseases associated with CD20 dysfunction include Ms4a1-related common variable immune deficiency.
