Product Specific Information

Antibody detects endogenous levels of total SOST.

Target Information

Sclerostin is a glycoprotein predominantly secreted by osteocytes and a member of the Cerberus/DAN family of BMP antagonists. It acts as an endogenous regulator of the canonical Wnt signaling pathway and inhibits bone homeostasis. Although mainly expressed in osteocytes, sclerostin is also found in bone, bone marrow, cartilage, kidney, and liver, and is produced by hypertrophic chondrocytes and cementocytes.
Sclerostin forms inhibitory complexes with LRP5 and LRP6, co-receptors essential for Wnt/beta-catenin signaling. It also interacts directly with LRP4 to facilitate inhibition of Wnt signaling and promotes osteoclast activity by increasing RANKL expression.
Deficiency or mutation of sclerostin leads to disorders such as sclerosteosis and van Buchem disease, characterized by excessive bone formation and hypermineralization.
