Product Specific Information

The eBio1D3 (1D3) monoclonal antibody reacts with mouse CD19, a 95 kDa transmembrane glycoprotein expressed by B cells during all stages of development except plasma cells. Follicular dendritic cells also express CD19. CD19 forms a complex with CD21, CD81, and MHC class II associated with the BCR, mediating signaling that induces tyrosine phosphorylation, calcium flux, and B cell proliferation.

• Applications Reported: Flow cytometric analysis.
• Applications Tested: Tested by flow cytometric analysis of mouse splenocytes; ≤ 0.25 µg/test recommended.
  A test is defined as antibody amount staining cells in 100 µL final volume (10^5–10^8 cells/test).
  Careful titration recommended for optimal performance.
• Alexa Fluor 561 emits at 575 nm and is suitable for spectral cytometers; multiplexable with PE and PE-eFluor 610.

Note: Internal testing shows Alexa Fluor 561 non-specifically stains B cells in Swiss Webster and SJL mice. Non-specific staining not observed in BALB/c or C57BL/6 mice.

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains, expressed on 100% of peripheral B cells. It may also be found on myeloid leukemia cells. CD19 serves as a critical regulator of normal and malignant B cell proliferation, functioning as a co-receptor with BCR to lower activation thresholds. Mutations cause hypogammaglobulinemia; overexpression leads to B cell hyperactivity.
