Product Specific Information
This 3DS223H monoclonal antibody recognizes human CD223, also known as Lymphocyte Activation Gene 3 (LAG-3).
LAG-3 is a 70-kDa surface glycoprotein of the Ig superfamily, homologous to CD4, and binds MHC class II with higher affinity than CD4. It plays a role in negative regulation of T cell activation and homeostatic proliferation.
LAG-3 is expressed on activated T cells (including regulatory T cells) and NK cells, with CD8+ T cells showing higher expression than CD4+ T cells. Coexpression of LAG-3 and CD49b may identify Type 1 regulatory T cells (Tr1).
Experimental Notes
- Recognizes a formaldehyde-fixed epitope.
- Reported for use in flow cytometric analysis.
- Pre-diluted and tested on normal human peripheral blood cells.
- Recommended usage: 5 µL (0.5 µg) per test (100 µL total volume).
- Compatible with violet (405 nm) laser-equipped cytometers.
Handling and Staining
- Use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize nonspecific polymer interactions.
- Protect from light to prevent photo-induced oxidation.
- Fixation: Samples may be stored in IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54) for up to 3 days at 4°C in the dark.
- Not compatible with methanol-based fixation.
