Target Information
The Pax genes are a family of transcription factors active in specific tissues during early embryonic development. Pax family members possess a DNA-binding domain encoded by the paired box, enabling them to regulate genetic programs.
Several Pax genes are associated with developmental mutations:
- PAX3: linked to Waardenburg syndrome
- PAX6: linked to Aniridia
- PAX2: linked to Wilms tumor
During embryogenesis, PAX2 is expressed in the developing kidney, particularly in condensing metanephric mesenchyme and early epithelial structures derived from mesenchyme. PAX2 expression decreases as the renal epithelium matures but persists in undifferentiated epithelium of Wilms tumors, correlating with proliferation of poorly differentiated epithelial cells. Expression of the Wilms tumor suppressor protein, WT1, coincides with down-regulation of PAX2, suggesting WT1 can repress PAX2 transcription.
