Target Information
Ubiquitin-mediated proteolysis requires the transfer of ubiquitin (Ub) to lysine groups on selected cellular proteins, promoting their degradation by the 26S proteasome. Ub-fusions are cleaved by Ub-specific processing proteases (UBps) or by the Ub-fusion degradation (UFD) pathway.
The UFD1 protein, first identified in S. cerevisiae, has a human homolog known as UFD1 or UFD1L. In vitro, UFD1 attenuates degradation of Ub-fusions with specific amino acid substitutions at Gly76 through selective multiubiquitination.
Mutations in the UFD1 gene are associated with CATCH22 syndrome, characterized by cardiac defects, cleft palate, and hypocalcemia, suggesting a role of this pathway in developmental processes.
