Target Information
Biliverdin reductase (BLVR) is a ubiquitously expressed cytoplasmic oxidoreductase that catalyzes the reduction of biliverdin to bilirubin. The process begins with heme oxygenase converting heme to iron, carbon monoxide, and biliverdin. BLVR is unique for its dual pH/dual cofactor dependency, using NADH (pH 6.7) or NADPH (pH 8.7) as electron donors.
Two isoforms exist:
- BLVRA (biliverdin-IX alpha-reductase) – major component in adult human liver
- BLVRB (biliverdin-IX beta-reductase) – predominant in fetal liver
Bilirubin, the product of BLVR activity, functions as an antioxidant but also acts as a neurotoxin causing kernicterus. It inhibits kinase activity and enzymes such as protein kinase C and NADPH oxidase.
