Target Information

GRP78 is a 78 kDa glucose-regulated protein belonging to the HSP70 family. It resides in the endoplasmic reticulum (ER) and transiently associates with newly synthesized secretory and membrane proteins. GRP78 interacts with mutant and misfolded proteins, marking them for degradation. The conserved KDEL sequence at its C-terminus ensures ER retention via the KDEL receptor.

GRP78 plays a key role in protein folding, degradation of misfolded proteins, and maintenance of cell homeostasis. It prevents apoptosis and serves as a biomarker of hypoglycemia. GRP78 provides neuroprotective functions under oxidative stress and glutamate exposure. Reduced GRP78 levels are observed in Alzheimer’s Disease brains and in PS1 gene mutations. Induction of GRP78 is associated with antitumor drug resistance.
