Target Information
Perforin은 세포독성 과립의 주요 세포용해 단백질 중 하나로, 세포질 과립에 저장되어 방출됩니다. 이 단백질은 종양 및 바이러스 감염에 대한 면역 방어에 관여하며, 세포독성 림프구에 의해 매개됩니다.
Perforin은 555개의 아미노산으로 구성되며, 21 아미노산의 시그널 펩타이드를 포함하고 분자량은 약 70–75 kDa입니다. C9 단백질과 유사한 막관통 채널 형성 기전을 가지며, 두 단백질 간의 상동성이 보고되었습니다.
Perforin은 킬러 세포 계통에서만 발현되며, 헬퍼 T세포나 기타 종양 세포에서는 발현되지 않습니다. 이 단백질은 T세포 및 자연살해세포(NK cell)에 의한 세포용해의 핵심 효과 분자로 알려져 있습니다.
Perforin 유전자 결함은 유전성 혈구탐식 림프조직구증 2형(HPLH2)을 유발하며, 이는 영유아기에 나타나는 희귀하고 치명적인 상염색체 열성 질환입니다. 또한, 대체 스플라이싱으로 인해 여러 전사 변이체가 생성됩니다.
