Product Specific Information
This 7-517 monoclonal antibody reacts with human CD276 (B7-H3), a type I transmembrane protein expressed on dendritic cells and various non-hematopoietic cells such as osteoblasts, fibroblasts, and epithelial cells. CD276 expression can be induced on dendritic cells, monocytes, T, B, and NK cells.
CD276 functions as a co-stimulatory molecule binding to activated T cells, promoting proliferation and IFN-γ production.
- Applications Reported: Flow cytometric analysis.
- Applications Tested: Pre-titrated and tested for flow cytometric analysis of normal human monocyte-derived dendritic cells.
Use at 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
PerCP-eFluor® 710 emits at 710 nm and is excited with a 488 nm blue laser.
Use a 710/50 bandpass filter (alternative: 695/40 bandpass). Ensure instrument compatibility.
Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark.
Excitation: 488 nm

Target Information
CD276 (B7-H3) is a co-stimulatory molecule that induces proliferation of CD4⁺ and CD8⁺ T cells, enhances cytotoxic T-cell generation, and stimulates interferon gamma production. Expression is inducible by inflammatory cytokines and found in tissues such as heart, kidney, testis, and colon.
In humans, CD276 exists as two isoforms due to gene duplication and alternative splicing. It has potential therapeutic relevance in regulating cell-mediated immune responses to cancer, especially with anti-angiogenic therapy.
