Target Information
MLCK (also known as MYLK3) is a myosin regulatory light chain kinase that facilitates muscle contraction. It belongs to the Ser/Thr protein kinase family and phosphorylates myosin light chains (MLC), enabling myosin–actin interaction.
MLCK plays a key role in vascular permeability and tissue edema formation. In the nervous system, it regulates astrocytic process growth and neurotransmitter release at synapses. It is also involved in fibroblast apoptosis signaling pathways.
Both smooth muscle and non-muscle isozymes are widely expressed in adult and fetal tissues. The non-muscle isoform 2 is the dominant splice variant in various tissues. The Telokin isoform binds calmodulin and is expressed in multiple tissues. MLCK is likely down-regulated by phosphorylation and contains one fibronectin type III domain and nine immunoglobulin-like C2-type domains.
