Product Specific Information

This FL34Z3L monoclonal antibody reacts with human CCL4, also known as MIP-1 beta. CCL4 is a chemokine involved in leukocyte recruitment to inflammation sites. It preferentially attracts CD4+ T cells, while CCL3 (MIP-1 alpha) attracts CD8+ T cells. CCL4 also induces cytokine release, mast cell degranulation, and NK cell activation.

CCL4 signals through CCR1, CCR4, and CCR5 receptors. CCR5 acts as a co-receptor for HIV entry, and CCL4 binding can inhibit viral entry by blocking gp120-CCR5 interaction and promoting receptor internalization.

Applications Reported: Suitable for surface and intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated for intracellular staining of normal human peripheral blood monocytes at 5 µL (0.03 µg)/test (100 µL final volume). Recommended cell range: 10⁵–10⁸ cells/test.

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm when excited with the 488 nm blue laser. Recommended filter: 710/50 bandpass (alternative: 695/40).

Fixation:
Samples can be stored in IC Fixation Buffer ([Product #00-8222-49]) or 1-step Fix/Lyse Solution ([Product #00-5333-54]) for up to 3 days at 4°C in the dark with minimal impact on brightness and compensation efficiency. Clone-specific performance should be validated empirically.

Target Information

CCL4 (macrophage inflammatory protein 1-beta, MIP1B) belongs to the CC chemokine family. It is produced by macrophages upon endotoxin stimulation and recruits inflammatory cells to sites of inflammation. CCL4 and CCL3 regulate immune cell trafficking and activation against pathogens. CCL4 selectively attracts CD4+ lymphocytes, while CCL3 attracts CD8+ lymphocytes. Both are chemoattractants for B cells, eosinophils, and dendritic cells. The processed form of CCL4 down-modulates CCR5 receptor expression, inhibiting HIV-1 entry into T cells.
