Target Information

Golimumab is a human IgG1 kappa monoclonal antibody derived from genetically engineered mice immunized with human TNFα.
It binds and inhibits both soluble and transmembrane forms of human TNFα, which are associated with chronic inflammation.

Golimumab is indicated for use in adults:

1. As an adjunct to methotrexate in moderate to severe rheumatoid arthritis (RA)
2. Alone or with methotrexate in psoriatic arthritis (PsA)
3. As a single agent in ankylosing spondylitis (AS)
4. As a single agent in moderate to severe ulcerative colitis (UC) requiring chronic steroids or with inadequate response to prior therapy

In the U.S. and Canada, golimumab is marketed under the brand name Simponi®.
The FDA label includes a black box warning for serious infections and malignancy.
In children and adolescents, lymphoma and other malignancies have been reported.

As a human monoclonal antibody, golimumab inhibits TNFα binding to its receptors, thereby preventing leukocyte infiltration and pro-inflammatory cytokine secretion (e.g., IL-6, IL-8, G-CSF, GM-CSF).
In patients with chronic inflammatory conditions, decreased levels of ICAM-1, IL-6, CRP, MMP-3, and VEGF have been observed.
