Product Specific Information

Description:
The 145-2C11 monoclonal antibody reacts with mouse CD3e, a 20 kDa subunit of the TCR complex. CD3e, along with gamma and delta subunits, is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. Binding of 145-2C11 to TCR can induce activation, proliferation, or apoptosis depending on conditions. It is widely used as a phenotypic marker for mouse T cells.

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes at ≤1.0 µg/test (in 100 µL volume, 10⁵–10⁸ cells/test). Optimize by titration.

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
  • Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
    • Add 5 µL per sample before combining with cells.
    • For single-color controls: 5 µL per 100 µL sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology, utilizing nucleic acid dye structures engineered for optimal fluorescent performance with minimal spillover and spread.

Target Information

The CD3 complex transduces antigen-recognition signals into T-cell cytoplasm and regulates TCR surface expression. CD3 subunits (gamma, delta, epsilon, zeta) are structurally related members of the immunoglobulin superfamily. They associate with signal transduction molecules through tyrosine-based motifs. CD3 is expressed on most T cells (68–85%) but not on B or NK cells. It is critical for T-cell activation, growth, and survival. Mutations in CD3 genes can lead to CD3 immunodeficiency.
