Product Specific Information
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
TIM3 (Hepatitis A virus cellular receptor 2, HAVCR2, T-cell immunoglobulin, mucin-domain containing-3) is a 281-amino-acid, Type-1 Th1-specific cell surface glycoprotein expressed on terminally differentiated CD4+ Th1 and CD8+ Tc1 cells.
It consists of an IgV-like domain, a mucin-like domain in the extracellular region, and a conserved tyrosine phosphorylation motif in the cytoplasmic region.
TIM3 is involved in macrophage activation and autoimmune disease induction, down-regulates Th1-mediated immune responses, and promotes immune tolerance.
Pathological significance includes its role in Experimental Autoimmune Encephalomyelitis (EAE), a Th1-dependent autoimmune disease, where it enhances disease severity in mice.
