배경 (Background)
Human epidermal growth factor receptor 2 (HER2)는 막 단백질로서 EGF 수용체 패밀리에 속합니다. HER2는 세포 발달, 증식, 분화에 중요한 역할을 하며, 다양한 암(유방암, 전립선암, 난소암, 폐암 등)에서 악성화 및 예후 불량과 관련이 있습니다. HER2는 EGF, TGF-α, amphiregulin에 의해 직접 활성화되지 않으며, 이합체화(dimerization)에 의해 활성화됩니다. PTK6와의 상호작용은 내재적 키나아제 활성을 증가시키며, EGFR, ERBB3, ERBB4와 이합체를 형성합니다. 또한 PTGS2/COX-2 프로모터의 특정 서열(5'-TCAAATTC-3')에 결합해 전사를 활성화하며, CDKN1A 및 rRNA 유전자 전사 조절에도 관여합니다.
