Target Information
Immunophilins are a family of soluble cytosolic receptors capable of binding to immunosuppressant agents such as cyclosporin A (CsA) or FK506. Proteins that bind FK506 are called FK506 Binding Proteins (FKBPs), and those that bind cyclosporin A are called cyclophilins (CyP). Both CyP:CsA and FKBP:FK506 complexes inhibit calcineurin, a calcium/calmodulin-dependent phosphatase involved in T-cell activation, explaining their immunosuppressive mechanism.
Immunophilins function as peptidyl prolyl cis-trans-isomerases (PPIase), catalyzing protein folding steps. Within the cyclophilin family, CyPA (also known as CyP-18) is the most abundant and ubiquitous form, present in all vertebrate tissues including T-cells.
