The TRI-COLOR® conjugate is a tandem R-phycoerythrin (PE)-Cy5 conjugate that enables simultaneous multicolor labeling and detection of multiple targets using a single excitation source—the 488 nm line of the argon-ion laser.

Target Information

HLA-DR is a transmembrane glycoprotein composed of a 36 kDa alpha chain (DRA) and a 27 kDa beta chain (DRB).

• The DRA gene contains 5 exons:
  • Exon 1 encodes the leader peptide
  • Exons 2 and 3 encode extracellular domains
  • Exon 4 encodes the transmembrane domain and cytoplasmic tail
• DRA acts as the sole alpha chain for DRB1, DRB3, DRB4, and DRB5.
• The beta chain (DRB) contains polymorphisms that specify peptide binding specificities.
• Hundreds of DRB1 alleles are known and used for tissue typing in bone marrow and kidney transplantation.
• Expression: Primarily on antigen-presenting cells such as B lymphocytes, monocytes, macrophages, thymic epithelial cells, and activated T lymphocytes.
• Function: Human MHC class II molecules bind intracellularly processed peptides, present them to T-helper cells, and play a critical role in initiating immune responses.

HLA and MHC antibodies are important tools in Immunopeptidomics, aiding neoantigen identification and characterization via LC-MS/MS.
