Target Information

Sonic Hedgehog (SHH) is expressed during embryogenesis and plays a critical role in early developmental patterning, including ventral neural tube, limb axis, and somite formation.
The human SHH protein is the most similar to the Drosophila counterpart and is produced as a precursor that undergoes autocatalytic cleavage. The N-terminal fragment provides signaling activity, while the C-terminal fragment assists in processing and covalently attaches a cholesterol moiety to the N-terminal product, restricting diffusion within the embryo.
Defects in SHH or its pathway can cause holoprosencephaly (HPE), leading to forebrain and facial malformations, and may also contribute to VACTERL syndrome, characterized by multiple congenital abnormalities.
