Target Information
FOXO1 (FKHR, Forkhead Box O1) is a transcription factor in the forkhead homeotic gene family.
It acts as a coactivator or corepressor of nuclear receptor activity via the LXXLL motif.
FOXO1 association with PAX3 is implicated in alveolar rhabdomyosarcoma.
It plays roles in autophagy, myogenic differentiation, redox balance, and metabolic homeostasis under oxidative stress.
FOXO1 binds insulin response elements (IRE) and Daf-16 family binding elements (DBE).
It regulates osteoblast numbers, bone mass, and glucose metabolism, and suppresses RUNX2-mediated transcription.
In hepatocytes, FOXO1 promotes gluconeogenesis through activation of IGFBP1, G6PC, and PPCK1 genes.
FOXO1 acts downstream of CDK1, PKB/AKT1, and MST1 in regulation of cell death.
