Target Information

Glial cell line-derived neurotrophic factor (GDNF) plays key roles in the control of vertebrate neuron survival and differentiation.
A novel neurotrophic factor, neurturin (NTN), was cloned from human and mouse. Physiological responses to neurturin require receptor tyrosine kinase RET and a glycosylphosphatidylinositol-linked receptor NTNRα.
The cDNAs encoding NTNRα from various species have been cloned and termed GDNFR-b, Ret ligand 2 (RETL2), or TGF-β-related neurotrophic factor receptor 2 (TrnR2), recently nominated as GFR-α-2.
NTN binds to and forms a complex with GFR-α-2 and the Ret PTK, activating the RET receptor tyrosine kinase pathway.
Both NTN and GDNF activate MAP kinase and phosphatidylinositol 3-kinase pathways, playing critical roles in neuronal development.
Neurturin and GDNF define a new family of neurotrophic factors.
