Target Information
Tau는 주로 축삭에서 발견되는 신경 미세소관 결합 단백질로, tubulin 중합을 촉진하고 미세소관을 안정화하는 기능을 합니다. C-말단은 축삭 미세소관에, N-말단은 신경 세포막 구성 요소에 결합하여 세포 내 연결자(linker) 역할을 수행합니다.
과인산화된 Tau는 Alzheimer’s disease(AD) 뇌의 신경섬유 엉킴(neurofibrillary tangle)의 주요 구성 성분이며, 미세소관 결합 기능을 손상시켜 신경세포 퇴화를 유도합니다.
Tau의 인산화는 GSK-3β, PKA, cdk5, casein kinase II 등 다양한 serine/threonine kinase에 의해 조절되며, Pick’s disease, frontotemporal dementia, corticobasal degeneration, progressive supranuclear palsy 등 다양한 신경퇴행성 질환에서도 발견됩니다.
