Target Information
Amphiphysins, members of the BAR (Bin-Amphiphysin-Rvsp) protein superfamily, are key regulators of clathrin-mediated endocytosis of synaptic vesicles.
All members share a conserved N-terminal BAR domain and a C-terminal SH3 domain. Two isoforms act as a heterodimer and interact via the SH3 domain with dynamin, synaptojanin, and clathrin.
These complexes are essential for synaptic vesicle recycling at nerve terminals. Amphiphysins are also known as dominant autoantigens in paraneoplastic Stiff-Man Syndrome.
