Target Information
Accumulating evidence demonstrates that cytokine receptor signaling is negatively regulated by a family of Src homology 2 domain-containing adaptor molecules termed SOCS (Suppressor of Cytokine Signaling).
There are eight members of the SOCS family: SOCS-1, 2, 3, 4, 5, 6, 7, and CIS. Structurally, SOCS proteins consist of:
- N-terminal region of variable length and composition
- Central SH2 domain
- C-terminal SOCS box motif
SOCS proteins function as part of a classical negative feedback loop regulating cytokine signal transduction via STAT-induced transcriptional mechanisms. Transcription of SOCS genes occurs rapidly in response to cytokines, and SOCS family members inhibit signaling in distinct ways.
