Target Information
CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase (PTP) expressed in all nucleated hematopoietic cells, comprising approximately 10% of all surface proteins in lymphocytes. It is absent on non-hematopoietic cell lines and tissues.
CD45 glycoprotein is crucial for lymphocyte development and antigen signaling, regulating Src-family kinases. Multiple isoforms exist due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic domains.
CD45RA is an isoform with restricted expression among lymphoid cell subtypes. Isoforms differ in their ability to translocate into glycosphingolipid-enriched membrane domains, depending on cell type and physiological state.
CD45 regulates T- and B-cell receptor signaling, suppresses JAK kinases to modulate cytokine receptor signaling, and promotes cell survival through integrin-mediated pathways, apoptosis regulation, and immunological function modulation.
