Phosphoinositide-specific phospholipase C (PLC) plays a crucial role in receptor-mediated signal transduction through the generation of two second messengers, inositol 1,4,5-triphosphate and diacylglycerol from phosphatidylinositol 4,5-bisphosphate.
Mammalian PLC isozymes include PLC beta1, beta2, beta3, beta4, gamma1, gamma2, delta1, delta2, and PLCε.
PLC delta exists as four isoforms. PLC delta1 acts as a calcium signal amplifier and is activated by an atypical GTP-binding protein. It functions as an effector for transglutaminase II-mediated oxytocin receptor and alpha1B-adrenoreceptor signaling.
Mouse PLC delta1 is highly expressed in brain, heart, lung, and testis. Abnormal accumulation of PLC delta in Alzheimer’s disease brains suggests its involvement in AD pathology. PLC delta2 is markedly expressed in type II intestinal metaplasia and adenocarcinoma, and may serve as a marker of gastric malignant transformation.
