Product Specific Information

The TN3-19.12 monoclonal antibody reacts with mouse, rat, and rabbit TNF alpha, but not with human TNF alpha. It is a neutralizing antibody. TNF alpha (17 kDa) is produced by macrophages, monocytes, neutrophils, CD4+ T cells, and NK cells, and also exists as a 26 kDa membrane-bound form. It plays an essential role in immune regulation and cytolytic activity.

Applications Reported:
Used for intracellular staining (flow cytometry), cytokine neutralization, ELISPOT, IVCCA, and ELISA.
Fluorochrome-conjugated TN3-19.12 is recommended for flow cytometry, and Functional Grade purified TN3-19.12 (Product #16-7423-81) is recommended for functional assays.
For ELISPOT capture, clone 1F3F3D4 is suggested.

Applications Tested:
Validated for intracellular staining and flow cytometric analysis at ≤0.25 µg/test (in 100 µL final volume). Cell number may vary (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal results.

Target Information

TNF alpha is a multifunctional proinflammatory cytokine of the TNF superfamily, secreted mainly by macrophages. It binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR receptors and regulates immune responses, apoptosis, lipid metabolism, and coagulation.
It exists as multimeric forms but appears as a 17 kDa band in SDS-PAGE. TNF alpha induces tumor necrosis in vivo and plays roles in bacterial, viral, parasitic, and fungal immune responses. It is implicated in autoimmune diseases, insulin resistance, and cancer.
