Target Information
Spleen tyrosine kinase (SYK), along with ZAP70, are cytoplasmic tyrosine kinases that serve as potential drug targets for allergic disorders and autoimmune diseases. SYK may also play a role in cancer. Upon B cell antigen receptor (BCR) or high-affinity IgE receptor (FcepsilonRI) activation, Syk translocates to the plasma membrane and phosphorylates downstream adaptor proteins, initiating signaling pathways such as calcium mobilization, cytoskeleton remodeling, and transcription of specific genes. Syk binds receptor assemblies via SH2 domain interactions with ITAM motifs phosphorylated by Src-family kinases, which also activate Syk through phosphorylation of its activation loop.
