Target Information
CD31 (platelet endothelial cell adhesion molecule-1, PECAM-1) acts as an inhibitory coreceptor regulating T and B cell signaling via dual immunoreceptor tyrosine-based inhibitory motifs (ITIMs). These motifs, upon phosphorylation, provide docking sites for protein-tyrosine phosphatases.
CD31 is ubiquitously expressed within the vascular compartment, primarily at cell junctions. Its N-terminal Ig-like domain mediates homophilic binding, essential for cell-cell interactions.
CD31 plays crucial roles in:
- Integrin-mediated cell adhesion
- Transendothelial migration
- Angiogenesis and apoptosis
- Negative regulation of immune receptor signaling
- Autoimmunity and macrophage phagocytosis
